Gene Expression Profiling of Glioblastoma Using cDNA Microarray

Abstract
Objective: The complementary DNA(cDNA) microarray was well known for new genomic large-scale screening techniques of gene expression profiling in brain tumors. The purpose of this study is to report of differential expression of gene at glioblastoma(GBM) using cDNA microarray.

Methods: GBM and non-tumorous brain tissues were obtained during surgery. Total RNA was isolated from each specimen and then we carried out gene expression profiling of 5 GBM tissues and non-tumorous brain tissues using cDNA microarray.
After that, to verify the reproducibility of expression genes, we performed semiquantitative reverse transcriptase-polymerase chain reaction(RT-PCR) using the same RNA used for microarray analysis.

Results: The analysis of gene expression of GBM and non-tumorous brain tissues showed up-regulated in thirty genes and down-regulated in twenty-four genes in the GBM comparing to non-tumorous brain tissues. The results of genes in RT-PCR analysis from each specimen were mostly consistent with those from the cDNA microarray study.

Conclusion: These results suggests that differential expression of gene may be useful to comprehend the molecular mechanism in GBM and to novel diagnostic marker. cDNA microarray study also is a new method for the identification of genes potentially involved in tumorigenetic pathways of GBM.
